Overview
Direct lysis** of tissue samples with Proteinase K (at 52 °C) can be performed on the robotic platform, avoiding mechanical homogenization. The lysis efficiency can be dramatically advanced by permanent shaking of the 96 well plate during the lysis step. The lysate is automatically mixed with binding buffer to adjust the optimal binding conditions and the DNA is bound to the membrane of the filter plate under non-chaotropic conditions. Contaminants are removed by washing twice and the ultra-pure DNA is eluted in a low-salt buffer. The purified genomic DNA is of high quality and can be used for a broad panel of subsequent downstream applications:

**For difficult to lyse samples, to reduce processing time or to prevent the risk of any clogging of the tips or needles of the workstation a customized protocol can be prepared including e.g. a homogenization step of the lysate on a mixer mill using Zirconia Beads. DNA from different stickleback eggs and spines was isolated using the Invisorb DNA Tissue HTS 96 Kit on the Tecan Freedom Evo 200. Purified DNA was used as template from different samples obtained with the procedure and DNA Taq polymerase from STRATEC Molecular. The figure shows amplified PCR products from Multiplex PCR using 5 microsatellites.
